Cytochalasin D (Zygosporin A) is a potent inhibitor of actin polymerization that can be derived from fungi. Cytochalasin D has cell-permeable activity. Cytochalasin D inhibits the interaction of G-actin-cofilin by binding to G-actin. Cytochalasin D also inhibits the binding of cofilin to F-actin and reduces the rate of actin polymerization and depolymerization in living cells. Cytochalasin D reduces the release of exosomes, which in turn reduces the amount of survivin in the tumor environment. Cytochalasin D induces phosphorylation of Yap, preventing its nuclear translocation and retention in the cytoplasm.
